Novel PEGylation and purification strategies for the development of biopharmaceutical products

Abstract
This work presents alternative and novel PEGylation reaction and purification strategies to develop biopharmaceutical products using three PEGylated proteins as models: PEGfilgrastim, PEGRNase A and PEGlysozyme. These strategies are presented as improvements of traditional PEGylation reactions schemes that involve sequential reaction and purification steps. The present dissertation is divided in five chapters. The first chapter serves as an introduction of the topic giving the general background regarding PEGylation as a technique to develop long- acting therapeutics and the methodologies used for its analysis. The results of the experiments herein proposed are presented in Chapter 2, Chapter 3 and Chapter 4, included in published or submitted scientific papers. The general conclusions of the dissertation are presented in Chapter 5, including perspectives for further studies in the same work line.
