Maternal health status is known to influence the immunological composition of colostrum. Therefore, this study aimed to evaluate the effects of maternal obesity on the antibody profile of colostrum, particularly focusing on vaccination-derived antibody responses, the presence of autoantibodies, and their functional capacity. For this, blood and colostrum samples from postpartum women were collected. ELISA assays were used to quantify antigen-specific IgG and IgA antibodies against SARS-CoV-2 and Tdap vaccine antigens, and to detect autoantibodies targeting mitochondrial DNA (mtDNA) and histone H3. Antibody-Dependent Cellular Phagocytosis (ADCP) was assessed using neutrophils and E. coli bioparticles to evaluate the functional effector capacity of colostrum and blood antibodies. Lean mothers exhibited higher vaccine-specific IgA responses, particularly against SARS-CoV-2. Anti-H3 autoantibodies were detected in both tissues but were lower in the colostrum of obese mothers. Finally, colostrum antibodies showed a reduced phagocytic activity compared to plasma, with a trend towards a greater functionality in samples from lean mothers.
